基于ISSR分子标记的江西省7个江西杜鹃自然居群的遗传多样性分析
Genetic diversity analysis of seven natural populations of Rhododendron kiangsiense in Jiangxi Province based on ISSR molecular markers

摘要

采用简单序列重复区间(ISSR)分子标记技术对江西省7个江西杜鹃(Rhododendron kiangsiense Fang)自然居群的遗传多样性进行系统分析,并根据研究结果提出针对性管护策略。结果表明:优选的3条引物(UBC825、UBC826、UBC827)在7个居群49株样株的DNA中共扩增出44个多态位点,多态位点百分率为86.3%。供试居群中,宜春市袁州区明月山居群的多态位点百分率最高(56.9%),吉安市遂川县南风面居群的多态位点百分率次之(33.3%),其余居群的多态位点百分率均低于30%。供试居群的观测等位基因数、有效等位基因数、Nei's基因多样性指数和Shannons信息指数分别为1.862 7、1.357 7、0.228 0和0.360 0,总遗传多样性指数为0.205 3,居群内遗传多样性指数为0.082 1,遗传分化系数为0.600 0,基因流为0.333 3。吉安市井冈山市水口居群与吉安市遂川县南风面居群间的遗传相似系数最大(0.946 4)、遗传距离最小(0.055 0),萍乡市芦溪县武功山居群与吉安市井冈山市龙潭居群间的遗传相似系数最小(0.745 1)、遗传距离最大(0.294 3)。个体间和居群间的聚类分析结果显示:吉安市遂川县南风面居群与吉安市井冈山市的水口居群、龙潭居群和八面山居群的亲缘关系较近,且这4个居群与吉安市安福县羊狮幕居群、萍乡市芦溪县武功山居群和宜春市袁州区明月山居群的亲缘关系较远。综上,江西省这7个江西杜鹃自然居群的遗传多样性较低,居群间的遗传分化较大、基因交流较弱,居群内的遗传变异也较低。针对江西杜鹃自然居群的生存现状和遗传多样性特征,建议采取就地保护、人工授粉和基因流管理、迁地保护和野外回归以及加强监测与研究等管护策略。

 

Abstract

The genetic diversity of seven natural populations of Rhododendron kiangsiense Fang in Jiangxi Province was systematically analyzed by using inter-simple sequence repeat (ISSR) molecular marker technology, and targeted conservation and management strategies were proposed based on the research results. The results show that the three selected primers (UBC825, UBC826, and UBC827) amplify a total of 44 polymorphic loci from the DNAs of 49 individuals across the seven populations, with the percentage of polymorphic loci reaching 86.3%. Among the test populations, the population of Mingyue Mountain, Yuanzhou District, Yichun City, exhibits the highest percentage of polymorphic loci (56.9%), followed by the population of Nanfengmian, Suichuan County, Ji'an City (33.3%), while the percentages of polymorphic loci of remaining populations all below 30%. The numbers of observed alleles and effective alleles, Nei's gene diversity index, and Shannon's information index of the test populations are 1.862 7, 1.357 7, 0.228 0, and 0.360 0, respectively, the total genetic diversity index is 0.205 3, the genetic diversity index within populations is 0.082 1, the genetic differentiation coefficient is 0.600 0, and the gene flow is 0.333 3. The genetic similarity coefficient is the largest (0.946 4) and the genetic distance is the smallest (0.055 0) between the population of Shuikou, Jinggangshan City, Ji'an City and the population of Nanfengmian, Suichuan County, Ji'an City, whereas the genetic similarity coefficient is the smallest (0.745 1) and the genetic distance is the largest (0.294 3) between the population of Wugong Mountain, Luxi County, Pingxiang City and the population of Longtan, Jinggangshan City, Ji'an City. The clustering analysis results among individuals and populations reveal that the genetic relationships between the population of Nanfengmian, Suichuan County, Ji'an City and the populations of Shuikou, Longtan and Bamian Mountain, Jinggangshan City, Ji'an City are close, and those between these four populations and the population of Yangshimu, Anfu County, Ji'an City, the population of Wugong Mountain, Luxi County, Pingxiang City, and the population of Mingyue Mountain, Yuanzhou District, Yichun City are distant. In conclusion, the seven natural populations of R. kiangsiense in Jiangxi Province exhibit low genetic diversity, substantial genetic differentiation and weak gene flow among populations, and low genetic variation within populations. In view of the current survival status and genetic diversity characteristics of the natural populations of R. kiangsiense, it is recommended to implement conservation and management strategies, such as in situ conservation, artificial pollination and gene flow management, ex situ conservation and reintroduction, enhancing monitoring and research.
